He did not initially intend to be an actor; at the University of Puerto Rico, he studied biology in an effort to become a doctor. After various acting gigs, Nolasco moved to New York City, where he trained at the American British Dramatic Arts School. He has also studied at The Marjorie Ballentine Studio, where he was a classmate of future Prison Break co-star James Hiroyuki Liao. After moving to Los Angeles to pursue his acting career, he worked at the popular club the Conga Room.
Nolasco was a supporter of the presidential campaign of Barack Obama and made a guest appearance in Will.I.Am's music video, "Yes We Can".
He has been romantically linked to former Miss Universe Dayanara Torres. He is dating Jennifer Morrison.
